چکیده
Background: The evidence on berberine stimulating the secretion of GLP-1 in intestinal L cell has been studied. However, few research explored its role generating islet α cell. Our experiment aims to clarify mechanism promoting and cell, activating GLP-1R downstream molecules through endocrine paracrine ways, thus improving function β treating T2DM. Methods: After confirming that can lower blood glucose improve insulin resistance db/db mice, identity maintenance, proliferation apoptosis cells were detected by immunohistochemistry immunofluorescence. Then, activation GLP-1/GLP-1R/PKA signaling pathway was evaluated Elisa, Western blot PCR. Finally, this verified vitro experiments Min6 cells, STC-1 aTC1/6 cells. Results: Berberine ameliorates metabolism mice. Additionally, it also increases number enhances This process is closely related activate GLP-1R/PKA autocrine paracrine, increase expression molecule such as GLP-1, GLP-1R, PC1/3, PC2, PKA, Pdx1. In , phenomenon enhanced signal had observed, which confirmed results animal experiments. Conclusion: maintain normal

Disturbed α-Cell Function in Mice with β-Cell Specific Overexpression of Human Islet Amyloid Polypeptide
Exogenous administration of islet amyloid polypeptide (IAPP) has been shown to inhibit both insulin and glucagon secretion. This study examined alpha-cell function in mice with beta-cell specific overexpression of human IAPP (hIAPP) after an oral protein gavage (75 mg whey protein/mouse). Baseline glucagon levels were higher in transgenic mice (41 +/- 4.0 pg/mL, n = 6) than in wildtype animals ...
متن کاملIntra-islet endothelial cell and β-cell crosstalk: Implication for islet cell transplantation
The intra-islet microvasculature is a critical interface between the blood and islet endocrine cells governing a number of cellular and pathophysiological processes associated with the pancreatic tissue. A growing body of evidence indicates a strong functional and physical interdependency of β-cells with endothelial cells (ECs), the building blocks of islet microvasculature. Intra-islet ECs, ac...
متن کاملIslet-1 Is Essential for Pancreatic β-Cell Function
Islet-1 (Isl-1) is essential for the survival and ensuing differentiation of pancreatic endocrine progenitors. Isl-1 remains expressed in all adult pancreatic endocrine lineages; however, its specific function in the postnatal pancreas is unclear. Here we determine whether Isl-1 plays a distinct role in the postnatal β-cell by performing physiological and morphometric analyses of a tamoxifen-in...
متن کاملRestricted islet-cell reactive T cell repertoire of early pancreatic islet infiltrates in NOD mice.
The mechanisms responsible for initiating autoimmune diabetes remain obscure. Here, we describe a method for identifying both the alpha- and beta-chains of the T cell receptor (TCR) from individual pancreatic islet-infiltrating T cells at the earliest stages of disease in nonobese diabetic mice (NOD). Analysis of the TCR repertoire of these early islet infiltrates reveals enrichment for a small...
متن کاملGlucose and Inflammation Control Islet Vascular Density and β-Cell Function in NOD Mice
OBJECTIVE β-Cell and islet endothelial cell destruction occurs during the progression of type 1 diabetes, but, paradoxically, β-cell proliferation is increased during this period. Altered glucose tolerance may affect β-cell mass and its association with endothelial cells. Our objective was to study the effects of glucose and inflammation on islet vascularity and on β function, mass, and insulin...
